Alpha - 1 - Antitrypsin - Human Leukocyte Elastase Complexes in Blood : Quantification by an Enzyme - Linked Differential Antibody
نویسنده
چکیده
An enzyme-linked immunosorbent assay (ELISA) has been developed for the quantification of cr,-antitrypsin-human leukocyte elastase (a,AT-E) complexes. In the ELISA, the a,AT-E complex is bound to a surface by rabbit antileukocyte elastase antibody, and the inhibitor-proteinase cornplex is quantified by a second antibody, rabbit anti-a1antitrypsin F(ab’)2. labeled with alkaline phosphatase. a1AT-E complexes were detected when a final concentration of 2.2 nrnole/liter of leukocyte elastase was added to plasma. The concentration of these complexes increased with additional elastase. In clotting blood, a1AT-E cornplexes were generated in parallel with the conversion of ‘251-flbrinogen to fibrin, whereas a2-plasmin inhibitor-plasmm (a2Pl-P) complexes were not formed. An enzyme-linked immunosorbent assay (ELISA) has been developed for the quantification of alpha 1-antitrypsin-human leukocyte elastase (alpha 1AT-E) complexes. In the ELISA, the alpha 1AT-E complex is bound to a surface by rabbit antileukocyte elastase antibody, and the inhibitor-proteinase complex is quantified by a second antibody, rabbit anti-alpha 1-antitrypsin F(ab')2, labeled with alkalin...
متن کاملDestruction of articular cartilage by alpha 2 macroglobulin elastase complexes: role in rheumatoid arthritis.
OBJECTIVE Neutrophil elastase accounts for the ability of some fresh rheumatoid synovial fluids to degrade cartilage matrix in vitro. The aim of this study was to determine if enzyme activity could result from depletion of synovial fluid inhibitors or protection of the enzyme from inhibition. METHODS The ability of synovial fluids to inhibit porcine pancreatic elastase was investigated togeth...
متن کاملDevelopment of an enzyme-linked immunosorbent assay with a monoclonal antibody prepared against alpha 1-antitrypsin for diagnostic screening of inflammatory disorders.
A monoclonal antibody, designated A2a18b8, of IgG1 class prepared against human alpha 1-antitrypsin, cross-reacts with alpha 1-antitrypsin in the serum of rat and baboon, but not with alpha 1-antitrypsin in serum of rabbit, pig, hamster, guinea pig, dog, or turtle. We used A2a18b8 in an enzyme-linked immunosorbent assay (ELISA) developed for human alpha 1-antitrypsin. Preliminary ELISA screenin...
متن کاملCorrelation between erythrocyte CR1 reduction and other blood proteinase markers in patients with malignant and inflammatory disorders.
Erythrocyte CR1, a C3b/C4b-binding complement-regulatory protein, is sensitive to proteolysis in vitro. To test the hypothesis that in vivo erythrocyte CR1 reduction results from intravascular proteinase activities, we used enzyme-linked immunosorbent assays to measure gamma-crosslinked fibrin degradation products (D-dimers) as indicators of coagulation/fibrinolytic activity, and complexes of n...
متن کاملLimited proteolysis by macrophage elastase inactivates human alpha 1- proteinase inhibitor
Inflammatory mouse peritoneal macrophages secrete a metalloproteinase that is not inhibited by alpha 1-proteinase inhibitor. This proteinase, macrophage elastase, recognizes alpha 1-proteinase inhibitor with macrophage elastase does not involve a stable proteinase-inhibitor complex and results in the proteolytic removal of a peptide of apparent molecular weight 4,000-5,000 from the inhibitor. A...
